A common strategy for starting the room is to hookshot to the bottom-left chest, but this is slower than simply walking diagonally. The hookshot can still be used after the diagonal walk, for the final movement upward to the chest, but the time save is marginal, even when done optimally.
Disregarding the Stalfos enemies, the fastest way to route the four chests and then exit the room is to circle the room clockwise or counter-clockwise. The actual strategy you use, however, will necessarily be highly dependent on your execution preferences, particularly in dealing with the Stalfos.
The shooters in this room trigger whenever Link is performing an action where his sword would be visible. Some shooters in the room will not fire if too many objects/sprites are on the screen at once -- including the Stalfos, their drops, and existing shooter fireballs. The shooters in this room are "prioritized" by their y-position, so the bottom-right shooter will always be prioritized first if there are too many objects on screen.
Because of this, the upper-left shooter will never fire until at least one of the Stalfos enemies is defeated. This can be used to your advantage if routing the chests in clockwise order, as you can safely dash without any risk of triggering that shooter.
This approach to the room takes advantage of the fact that the leftmost stalfos will always stop at the shooter, allowing it to be consistently killed with an arrow after moving up-right from the door. The bottom stalfos will generally be within reach for a sword slash, leaving only the top one to track and kill using either an arrow (which will avoid triggering the shooters) or a slash/dash, as appropriate.
You'll need to walk both down and left after opening the second chest before dashing to the left -- attempting to dash form the far right side of the room will result in being hit by the top-right shooter.
If the top stalfos moves all the way to the far right of the room, it's possible to shoot an arrow past the edge of the chest to guarantee a kill, though optimally this is slightly slower than a slash.
